You definately want something with a good sample editor. Kontakt is probably the king of the mountain on that, but since you mention Wusikstation,,, you might have thiers in your toolbox already,,, and the Wusik editor is really slick. It's a good place to cut your teeth.

You need to be consistant in the way you play each note, need to decide the distance of the intervals you're going to sample, and you need to play the notes at consistant volumes, so that you have the tonal character of the samples to match the volocities you intend to map. You also need to set the loop points if you intend to have them sustain.
Personally,,, the type of samples I create are really more for sound effects compendiums, etc. For instruments, I usually buy samples, because there are so many good ones available, and offer pretty good VFM. That's not to discourage you from going ahead and jumping into sampling, though.
