I'd have to think a bit longer about new things, but the current things that most annoy me on a daily basis are..
 
In the track inspector the clip info tab doesn't shrink back the inspector like the track info and PC tabs do. ie. if  I have the inspector just showing the track info and not the bus also, clicking on the PC tab will resize the UI to use the PC. Click on it again when your done and it reverts to the previous state, TI tab is the same, but Clip tab opens but never closes. Always having to click on one of the other two tabs and then close that instead. A minor thing maybe, but it's really annoying and I would imagine a relatively simple fix as it works fine in X3, but somewhere along the line a Platinum update broke it.  I can't say when exactly because my first installed Platinum version was 2016.12 and it was already broken by then.
 
Lack of a solo button on pro-channel eq.
 
The undo bug - Edit>Undo doesn't show the last action, but the one before it. Again something that got broken along the way and repeatedly ignored.
 
New features that have come to mind while writing...
 
Console view folders - a user option to have the same effect on the console view when a track folder is closed.
 
Ability to reorder the synths in the synth rack, just for the sake of tidiness really.  On larger projects it would be nice to be able to see all of the instances of the same synth displayed adjacently rather than just in whatever order they were added to the project.
 
The browser could do with some love - things like the abilty to show the UI of the synth/fx as a thumbnail (whispers quietly........like in studio one).

The only thing I can think of at this time is please bring back the ability to add or delete notes within Staff View...I miss the pencil and eraser tools.....:)

4) Staff view improvements (Note duration toolbars and triplet handling)
5) Tempo track and time signatures on the main window

Wow, Meng... thanks for opening this up! I am in awe of the astute ideas posted above and can only agree without adding the same things over again. I will insert these:
 
Right now it seems that *only* the Mercury theme has the ability to change the track controls color. All other themes just change the color of the vertical bar at the far left in track view. I also want to change the color of the track marker ellipsis to bright yellow... the faded gray that is current gets visually lost. I have done this in track editor but then the Mercury style track color goes away and I am left with vertical bar color and yellow ellipses. I want both!
 
I would like a new DAW announcement for my birthday, April 6... clock's ticking, Meng!

I'd like to see something like AZController integrated in TDFKAS, instead of the current ACT. 
And better/easier controller integration for HW controllers.
Probably have more at a later time.

Before Cakewalks closure there was an opportunity to intergrate Overture (a stand-alone notation software) but it was rejected by Gibson. Perhaps this could go ahead if it is indeed practical. It could go a long way toward answering all the notation requests and bugs.
One of the oft voiced complaints is that features are added in incomplete forms. AudioSnap, Matrix and Step Sequencer could all do with enhancement.

Just a couple that I can think of off the top of my head:
1. More metering options (example: Peak to RMS to K-20, 14,12, Ebu-128, stereo field, phase, etc.)  This would be very helpful being built right into the DAW without needing third party plugins snagging up computer resources.
2. Placing plugin inserts as Pre or Post fader without having to use a send.

1. Studio One lets you have 1 midi file trigger multiple VSTi's.  ive seen workarounds using input echo and recording on fly but thats nonsense to me.  i just want 1 file to run through 2 VSTi's without having to arm this one and record that one, etc.  If there was a simple way to select the VSTi's input as being any midi track on the track view, that would be awesome.
 
2. Reaper has a really cool range slider for velocity dynamics in the midi editor.  when you select a range of midi notes and open the velocity tab, the slider lets you flatten or randomize the velocities of the selected notes on the fly.  it's very helpful and super quick to randomize things like hi-hats.  ive used the midi scripts/function that are currently available, but to me, they seem very inconsistent and i have to fiddle with them a million times just to get something descent when wanting to randomize note velocity.  having a simple dynamics slider is cut and dry in my opinion.
 
3. one bug i get all the time is when i save a track template that uses a drum map, it almost never ports over to a new project when i use that template.  i always have to go onto the track and pick the drum map again.  routing and everything else saves properly, but the map always changes. to some default in the map manager
