I have Melodyne Editor installed, do I need to select Melodyne option in BandLab Assistant checked in order for Melodyne to work? Thank you. 

No, you don't. It will install a trial if you do, and might mess up your current Melodyne if you do.

Hi vladasyn . . . all installs and authorizations for Melodyne Editor are done from the Celemony site.
If it's not working for you in your Cakewalk by BandLab install, you probably need to find where the Melodyne VST is . . . . and add that path to the VST scan in Cakewalk preferences.
