I seem to recall it requires a registry hack? Is there a preferred method for moving this content off of C (I think it's in Cakewalk Content/) and onto another drive? I don't really use DimPro anymore (or Beatscape for that matter, of which there's also a huge folder of samples), but I don't want to break it for old projects.

I believe it is much easier and safer to use mklink, a command line utility, to relocate data. There is no need to mess with the registry at all. I know there are quite a few post containing examples of how to move data around.

The best info on this is by Steve (scook) himself which I've put to good use in just the last few days.
