Hi all,
 
I recently installed 3 samsung 850 evo drives.
Win7, audio, softsynths/samples.
I used the Samsung magician software to migrate.
I'm noticing some audio glitching lately. 
 
Are there any tweaks I should do for these drives ?

The Samsung Magician that comes with the Evo drives will give you the best performance options.
I wish other SSDs came similar tool sets (maybe some do, the Mushkin I picked up for my other system didn't)
As for tweaks you have to decide what you want more performance vs reliability - SM does pretty good job at explaining the options and giving control over them (or if not control explaining what to tweak - been a while since I last played with it so don't remember 100%).
I've opted for performance and I haven't seen any app or setting tweak that appears to do more than what S.M. does or recommends.  
 
SM has Rapid Mode which seemed pretty impressive.  It can give you a nice performance boost but it can only accelerate one drive in the system. 
 
You say your getting audio glitching but you don't say how you have the 3 drives connected and that may be a factor.
Or it could totally be something else.
I have an Evo 1TB SSD as my C: drive on my laptop.  If I don't turn off my WIFI I get glitching and audio engine crashes.

I (and most of the techie's from posts on computer forums) do not use the Samsung Magician tweaks.  I tried them and they screwed up my system, including power configuration.  Most only use the Samsung Magician to check for firmware updates.   
 
I have two Samsung SSD's, an 840 EVO 500GB and an 850 EVO 500GB.  The 840 is for Win 7 OS and the 850 is for Win 10 OS and Sonar.  I keep my Sonar projects on a separate conventional hard drive.

Yea, I noticed the power configuration getting changed. 
I also notice that Magician and some other Samsung app is always running (task mgr).
 
Ok, well, I think I'm gonna swap out my audio drive back to the standard 7200 HDD.
I don't really notice anything beneficial with an SSD as an audio drive. 
 
 

Most SSDs require AHCI mode enabled in the BIOS. Additionally, there are several do/don'ts with them. They excel at reading, and not exceptional with writing, so the best use is on a drive that will (primarily) have static information such as the O/S drive itself.
 
Settings internal to the O/S for an SSD are static, so the "Magician running" is a bit concerning. If you run "msconfig.exe" you can disable programs which run at startup on your machine.
 
If you google "win7 tweaks for SSD" the top several links all have walk-throughs of things to do/not do with them, but I assume that Samsung's Magician *should* have done all of these already. Why it loads at startup I cannot speak to - all of the things you require are one-time only.

From what I recall Samsung's Magician default settings are very conservative.
Even if you don't use it check to see what it would do to optimize performance - these were the same setting recommendations I saw being made out there when I got my SSD. I don't think that they have changed much.
 
You need to find what the optimal settings are for your motherboard's controllers.
Search for articles, forums or forum threads about your ASUS P8P67 and see what the 'experts' recommend.
For example ACHI probably should be enabled but that is not universal and may not be the case for your system (on mine for example it has to be turned off).

I did the 'disable indexing'. (and, whatever Samsung Magician does).  
I’m in AHCI mode. Trim enabled.
"Disable system restore"? Hmm. Don’t think that’s a good idea. I use it often.
Turned off drive indexing on my audio drive. Should I also do C: drive?
Power settings. Yes.
 
"Disable Windows Search and Superfetch"?
Is this one important? 
 
But, most of the other stuff especially registry hacks seems a bit much???
Are they safe and important?

I went rhu a VERY tedious process of moving event logs for the most active sets to a different drive.
