can an external samples drive be USB?

i know that an external eSATA drive is recommenced for the audio drive but can the samples drive just be USB based?

are the samples read from the hard drive when recording through Sonar and recorded onto the audio drive?  or are the samples first loaded into RAM and then the project recorded?

    YES.

For quite some time, before I built my new DAW, I ran my EWQL and Kontakt samples from an external USB drive. I also ran a few other sample based application that way as well.

As long as the Application loads the samples into RAM memory you can have them anywhere.  If the application uses streaming samples that might be a different story, but none of mine are streamers so the USB works perfectly fine.
