Sounds like you need to over write the base sample with your new sound - ( same name and location ). Then bounce to clips. Then call it a done deal and go back and correct the base sample. What you seem to be asking for is a sample replacement tool and that's essentially what Bit Flipper is recommending via a sampler. If it's a drum / percussion sound then Battery is a very good tool for this. I imagine drumagog is very good for this too.

I think it is possible to replace the base sample, but you must do it 32 bit floating point i think. Else it wont work
