First you need to figure out if it's a MIDI problem or an Audio problem. TTS-1 has an audition button at the bottom of each channel that will demo the selected instrument. Insert it with separate MIDI and synth audio tracks so you can treack MIDI and audio I.O and meter activity separately. If you see meter activity in the TSS-1 GUI, but don't hear anything when clicking the audition buttons, follow the audio path from the audio track input assignment all the way to your monitors. If you hear the audition, but TTS-1 doesn't respond to MIDI, follow the path backwards form the Output assignment of the track to your controller (or the MIDI events in the clip if playing back existing MIDI).
 
If the problem is that synths arent responding to your MIDI controller, my first guess would be that you disabled Always Echo Current MIDI Track in preferences, and are neglecting to enable the Input Echo button on MIDI/Instrument tracks so that live MIDI input gets echoed to the synth.
